High strength and endurance, maybe agility of 7 for Slayer later on.

Although the my advice is go straight to Sloan and get a Sledgehammer.

As for joining CL, you have to get to Benny first to get to them.

high endurance and high strength ofcourse.. one my last playthrough I also kind of had a melee build, and having a Science skill of 50 was pretty handy, because chems such like Slasher can be really useful. The Chemist perk is also pretty cool

If you like Turbo..
Spoiler
you should go to Red Rock Canyon and complete Aba Daba Honeymoon and ask for a recipe, Diane will give you the recipe to make Turbo

High strength, high endurance, highish agility if you want it (+ to run speed is useful)

I went low charisma, low perception, high intelligence and good luck but that's my preference for the character.

My guy was basically pure melee until he maxed it, repair, lockpick, sneak and got unarmed up to a high enough place for the perks. Now he varies between medicine, unarmed, guns and science.
The only time he uses ranged weapons is if he's a) unable to get to melee (some enemies are high up or w/e) or if he's pretending to be an NCR ranger...but normally he just bashes them.
Join Caesar's Legion by following the main quest, get to Vegas asap then after meeting Benny you'll be approached by a representative of Caesar.

I recommend stopping off at Camp Searchlight and obtaining Knock Knock as well. It's a fairly easy quest and the axe is well worth it.

False, Agility doesn't increase running speed. But it's definately worth having an Agility of 7 (or 6 + Implant) for the Slayer perk. Also that thing about Knock-Knock should have a spoiler thingy.
Spoiler
Knock-Knock is indeed incredibly useful. When fighting enemies that are carrying any kind of weapon Knock-Knock has a chance of knocking the guns out of their hands. (this includes unarmed weapons, I've knocked power fists out of enemies' hands myself)

EDIT: Consider the Heavy Handed trait. It's very important wether you decide to focus on critical chance or not. The Heavy Handed trait is incredibly useful IF it's used correctly. If you focus on crits and don't take the trait, you can be just as effective.

Light armor and Travel Light for the +10% run speed.
Reinforced Leather Armor, two ranks of Toughness, and NEMEAN, and headgear of your choosing will equate to at least 20 DT

One thing you have to look out for is the Piercing Strike perk, if you're going pure melee. It applies to both melee and unarmed but the requirement is a fairly high unarmed skill, which means you basically have to sacrifice another potential skill. It is very much worth it though, even necessary for melee to be useful later in the game. You will also probably want either a very high DT or get really good at sneaking to take on ranged enemies. Silent Running is vital if you go the stealthy approach. Too many times I've had someone turn around and spot me right before I get into VATS range because I took too long sneaking up to them.
