I am still friendly with Brotherhood, Minutemen and Railroad,

Got Quest" Defend the Castle" marked already, but it won't trigger.

Did hundreds of Quests for Preston, think I got enough settlements.

Simply cannot continue.

PAM keeps giving me new Quests for the raildroad, can't talk to Desdemona, though.

What am I doing wrong?

Help would be highly appreciated !

Railroad ending can't be complete once you become enemies with the Institute.

If you can't get doctor Li back then you can't complete BoS ending either.

Institute ending is blocked off since you have angered them.

Not sure but listen to the radio after the having 8 settlements and speak to Garvey should have initiate the quest. They should tell you where the attack coming from at the castle.

seems like an endless loop.

I was a victim of the bug in the Concord quest, probably did too much other

quests for BoS and Railroad ( Did Institute Quest for Railroad),

By the way,Is there a way to get back into the Institute after I got hostile with them?

No, you can't get back with any faction once you have become hostile with them, due to its ties to the other factions which required infiltration, siding against the institute too soon would basically lock you out of both BoS and Railroad. Basically, you're forced to side with the Minutemen.

Maybe it's a bug, if you're on PC, you can reset the quest with console command, if you're on console, you're out of luck and may have to use an earlier save.
