Yes, but I haven't tried it myself. Maybe there is someone who did. I have seen them ingame. I had one with two daggers though.
You can create any type of character combination you want. So, yes you can.
I believe any stamina based weapon skills work perfectly fine from stealth. You will notice no weapon offers stealth enhancements (I believe). The weapon is totally up to you.

I was just looking into doing this for my Skyrim game. In that game, there are perks you can get that would allow daggers to do 15x damage from sneaking backstab but Two Handers have no such perk. So if you had a 9 damage dagger, it would do 45 damage from hiding... but if you had a 45 damage two hander, you'd do the same damage AND be able to do that damage all day long whether you were hiding or not. The question then becomes, how much harder is it to get a 45 damage two hander versus a 9 damage dagger (and how hard is it to get the dagger's x15 sneak attack bonus perk).
In ESO, I don't THINK that they have an explicit dagger bonus to backstabbing. If that's the case, then there doesn't seem to be any real disadvantage to using a two hander, except the intrinsic differences (slower attack speed, longer reach possibly, etc). Come to think of it, though, there are some Dual Wield skills that give different bonuses if you have a sword or axe or dagger equipped, I believe. I don't know the two hand weapon skill lines well enough to say if they offer anything that would be particularly useful for an assassin.
