I suspect we might see more of The Abbey of the Road, the Christian group who sends out missionaries like the one we meet in Point Lookout.
Going by her logs, Boston is the closest place to the abbey that we know of.
We also might see Desmond Lockheart, and other players of "the great game". Desmond said he was going north at the end of PL.
-Raiders, or some other name for a faction of anarchic groups that go around killing.
-Brotherhood of Steel, they've been in every Fallout game so far so it's a safe bet they'll be back. Whether it'll still be Lyons' Pride (Boston is quite close to DC, after all) or the Midwest Brotherhood is anyone's guess. Maybe even both!
-The Insitute, since it's in Boston.
-The Railroad, assuming the Insitute still treats synthetics the way they did in Fallout 3.
-Each individual settlement is likely to be its own faction, unless the entire region was united, but that's not likely.
-Maybe the Enclave? I think by now they're a spent force, but who knows, maybe they'll swoop back in.
-I doubt we'll see the NCR, the Legion, or any other NV faction save the Brotherhood. The east coast is quite a way off from the Mojave. Maybe the odd traveller who's been there to drops tidbits of info.
-Some sort of Slaver faction. Slavery's good money, so I'm sure we'll see some in the region.
Well i personally would love The Enclave and The Brotherhood to have a presence in the game as they're my favorite factions for a multitude of reasons. Also learning more about the secretive Institute is one of my most anticipated aspects of the game.
Honestly i'm just glad that the NCR will most likely not have a presence seen as though they are are west coast based faction i believe. I hate the NCR, they're so boring imo.
Instead of slavers i would rather have some 'android catchers' mercenary's hired by the Institute to do their dirty work.
Maybe if you have the Android perk they will come after you to .

I have no issue with the BoS coming back. I'd at least like them to feature in some way (even if it's comments from NPCs) considering it is probably set after FO3.
That said, I hope there are more than two factions. Two factions end up becoming a binary issue between good and bad. Bethesda did a slightly better job in Skyrim with the Stormcloaks/Imperials and neither side fitting into good or bad. But I think having the choice to side with three or more factions gives the factions and player choices some complexity.

Any group who uses power armor will be the group I go with.
Unless the NCR somehow manage to rear their ugly heads with their crappy salvaged power armor than yeah, same with me.
That's the spirit, loyalty to the ones with the toys.
Honestly, since Lyons Brotherhood (LB) most likely won in Fallout 3 it'd make sense for them to turn their sights elsewhere as the Capital Wasteland is getting cleaned up so I wouldn't mind them being in Fallout 4. It's not like Fallout 3 where BOS were forced to the east coast for no other reason than iconicism(?). LB is relatively close to Boston. Boston which has a faction with really advanced technology. Why wouldn't LB show an interest in it? Besides, super mutant refugees and Enclave Remnants might be holed up there and LB in their crusade might be an invading force who will stop at nothing until they crush their enemies.
I'd prefer it if this new power armor factions is in fact that, new.
But if LB shows up then the only demand I really have is that they are not called BOS. Because they're not BOS. They ought to be called Lyons Brotherhood or just The Brotherhood or something but they should absolutely 'not' be called Brotherhood Of Steel. There is already a huge misconception(?) among Fallout 3/NV fans about what BOS actually are like and it doesn't help the lore one little bit to call two factions so drastically different from one another the exact same thing. Especially not when one has a feasible replacement name already.
So yeah, super mutant refugees (hopefully they won't be automatically hostile), Enclave Remnants and Lyons Brotherhood are probably going to make a return.
Some new ones would be good, but i don't mind The BOS returning or the Enclave in some form.
They went east to recover tech from the D.C., and to investigate rumors of a new kind of super mutant.
End of Fallout 3 the Brotherhood there's become more of a governing body, it could have formed it's own power base I guess. Maybe the outcasts could travel north to escape them and be the BoS forces there?

Super Mutants should never have been at the east coast in the first place. They too were shoved in there for the sake of iconicism(is that even a word?).
And tech? It doesn't make any sense to send a whole chapter across an entire continent to look for tech when there is an abundance of tech still around the west coast.
But whatever. BOS went to the east because they were all high huffing paint. I don't even care any more. Sadly it is canon and there is nothing I can do about it.
My point was that at least 'this time' it'd make sense for them to show up in a game because it is close-by and has a faction that could be of interest to them.
And while I long for the day we have a Fallout game without stupid space armor I'd rather the involvement of factions to make sense over my petty disdain for space armors.
Well it's explained ingame and considering that they are not really like West coast super mutants....
I think the Brotherhood needs to be in the game, but as a small side-faction. Perhaps even smaller than it was in New Vegas.
Enclave would be a neat footnote and rumor, but they need to be done. I hope they come up with new main factions.

1.The FEV was orginally researched by West Tek but the goverment choose to put it under military control.Vault Tech with their close goverment ties could defintly get samples.The tought that they may have wanted so of it to remain in a vault is not that far stretched.
2.The Pentagon,the fact that itis the former capital?Accoridng to them in the game they only noticed that the Super Mutants were there when they arrived.
I'd love to see the Midwestern BoS somewhere.
