As much as I personally would want to be able to continue after endgame, I'd have to vote no, mostly for the sake of the devs. It would be a programming nightmare, I'm sure, considering all the various endings.
However, after seeing threads like this (many, many times), I think I may have a good idea for a compromise. That is, New Game+. What happens is that the game is over, but you start a new game where you can pick up where you left off with your challenges (and keep the challenge perks you acquired along the way). I think this would have the effect of encouraging people to replay the game using very different builds and approaches. It really becomes a fresh game if you go from a stealthy sniper, to an in-your-face melee fighter, to a smart talking, fast-dealing pistoleer. So, in a way, you get to continue your game. However, it's more like a "reincarnation" than an "afterlife".
The challenges might have to be tweaked a little, so that each one has to be completed within a single game. In other words, "I Dismember You" would required 1000 dismemberments in a single game, not spread across a few games. I'm sure it's easier to carry over character data over to a new game than to deal with post ending game play. With that in mind, I would also like to see more challenges, as well as more difficult challenges.
That's just my two cents.
