I just unlocked the Local Leader Perk but can't seem to figure out how to set up the supply lines.

Supply lines and moving settlers from one settlement to another work about the same except for pressing a different key/button. Look at the settler in the workshop menu and you'll see options for moving and assigning a supply line at the bottom of the screen. On PC, moving is "R" and supply line is "Q".
