I killed him once.... but i never went back to find out if he get's replaced. sadly that was 2 games ago, so i have no idea. but i do know that certain merchants will get replaced if killed, and i would assume he is one of those merchants.
I killed Belethor on my first run through and no one EVER took the shop back over. I was probably around level 10 or so and played that character to +50. The store would respawn food and that's about it.
Don't kill him. It's a major PITA without him there because he's the only one in Whiterun who buys everything.
Not always true. My mage has the speech perk that allows him to sell anything to anyone. It's really nice. Never thought about killing Belethor though even though I can't stand the guy. I hate that smug line he drools out every time I stop talking to him. I just want to turn around and punch him in the face.

If you kill Belethor, Skyrim will be a much less annoying place. While you're at it you might as well take out all of the characters voice-acted by that same guy. They are tainted by association.
