Do you have to complete the nights of the nine before you earn the pilgrams grace?

No, but if you have KOTN installed, you must have Zero Infamy to get Pilgrim's Grace. If you have KOTN and have infamy, you must visit all the shrines to clear that infamy first, and then visit all of them again in order to receive the power.
