One problem with playing any sort of Super Mutant-type character would be a severely limited weapon selection as their fingers will not fit the triggers of most Guns or EW. On the other hand, since SMs can have a STR in excess of the normal 10 cap you could carry multiple heavy weapons with no real capacity loss, especially if you also take the Heavyweight perk, however most of those have a 75 or 100 skill requirement so you'd be operating well below the norm for a while. Heavy-Armor melee tanks would be their specialty, as they would also have a really high END.
Ghouls wouldn't be able to raise STR, END, or AGI very high and would also have a hit to CHA if going by lore, however they'd get a bonus against Radiation and Poison effects as well as a high regeneration factor based on how irradiated they happened to be. Heavy weapons would mostly be out but Sneak and Survival would get bonuses, which would make them excellent Ranger-types.
The above is pretty bare-bones, but you get the idea. Would need a lot of fleshing-out to be properly playable, and I'm not sure it would be worthwhile given the likely high difficulty in interacting with the Human inhabitants of the Wastes.
Playing as a Super Mutant a lot of communities would shoot you on sight, while as a Ghoul they would be suspicious/paranoid towards you to say the least; in either case, if you somehow did manage to get them to talk instead of shooting there would have to be a whole bunch of specialized dialogue reflecting that race's inherent standing with that particular location.
As such, unless the developers go to great lengths in dialogue, NPC attitudes, and face generation it just won't work, as all you'll be doing is playing a green-(SM) or yellow- (Ghoul [Glowing One-class]) skinned human with a few 'cosmetic' traits slapped on and that would be utterly unsatisfactory to say the least.
