I'd say that Arthur Maxson is a certainty personally.
Well, I can't say I won't miss him. But, I can only hope there will be new characters for me to love and enjoy.
Damn, I was really hoping he'd be in it. This is a pretty sizable disappointment.
Well, maybe not the protagonist but I'd say his presence and the Brotherhood's would be indicative of the ultimate outcome of Fallout 3's events. Enclave loss

Not a big loss. Sure, I'd still love to have Erik in, but Three Dog's character was a little too over the top for my tastes. Mister New Vegas was like a breath of fresh air when I first heard him, hopefully FO4's DJ is reminiscent (not a clone) of New Vegas' DJ.
Good. Not that I don't like Three Dog, I'd just like to see new characters for a new game.
(and plot-wise, if the timeline is close to when Fallout 3 happens, I couldn't see him abandoning the "Good Fight" / the people of the Capitol Wasteland / his station. Wouldn't fit the character. Honestly, not even if it was a couple years after FO3.)
Glad to see Three Dog not returning, shame to not see Erik Todd Dellums return to voice at least one character. I had thought after his second role in Skyrim he would have to be a staple voice actor in a Bethesda game like Wes Johnson. Guess that's not the case.
Not entirely so. NPC wise, yes, his roles were limited. But he also voiced Super Mutants and Sentry Bots in Fallout 3, and once again the Dremora in Skyrim. He has always been good at those eccentric roles.
Regardless, I'm okay with him having less roles, but voicing unique NPCs. His talent really adds to what makes his characters so great like Sheogorath and Hermaeus Mora. I would hope he returns to voice another unique NPC in Fallout 4.

It'd make sense given that he went on and on about fighting the Good Fight in the Capital Wasteland. It wouldn't make any sense for him to pack up and head to Boston, even if Fallout 4 happens years after Fallout 3. His place is there at the GNR, and always will be.
You guys remember when Todd(3-dawg) said he had "permission" that more three dog may be on the way? Did he do something wrong and screwed himself out of a job. Why would he say that?
