In my eyes, the pro does not outweigh the cons.

Pro- rad resistence/health regen

Cons- voice (most sound pretty awful with a few strange exceptions), appearance (why go through all the character creation only to have them disfigured), reaction from NPCs (most people don't like ghouls and should react accordingly to you)

I'm sure there's more reasons not to but I can't really think of more reasons FOR ghoulification

It doesn't protect you from radiation it just restores your health when you get radiation, you still get radiation of course or it wouldn't work. Bethesda might of jumped the gun on the naming of that perk, it probably won't alter your appearance at all.
