Depending on the environment obviously.
For the main quest, I want a very gritty, dark, and emotional side to it. Personally as a roleplayer and immersion advocate, I want to feel attached AS A PLAYER to characters I'm supposed to feel attached for and also feel hatred towards the villains, or maybe sympathy. But I also want it to be dark and real. No butterflies and lollypops to brighten any mood, it needs to feel real and in the now.
For side quests, it really depends. If it's something like deliver something to somebody, it can really be anything it wants to be, but personally, I want it to be a little light as it's a regular story...unless it's a conflict. Conflicts should be dark by nature.

Towns and other quiet areas should have an emotional and cozy nature to it while large cities should have a busting and fast nature to it. But any normal settlement should have a lighter tone to it...unless that said settlement is inhabited by Glarthirs.
